1, Apr 2012Authors: Nakajima H, Ezaki Y, Nagai T, Yoshioka R, Imokawa G Abstract We recently reported that overexpression of the elastase NEP (neutral endopeptidase) by fibroblasts plays a pivotal role in the mechanism of UVB-induced skin wrinkling by degrading dermal elastic fibres. Since UVB does not penetrate to the dermis, we hypothesized that factors secreted by UVB-exposed keratinocytes in the epidermis trigger fibroblasts in the dermis to increase their expression of NEP which then degrades the elastic fibres. In the present study, we characterized the epithelial-mesenchymal interaction between keratinocytes and fibroblasts which leads to increased expression of NEP

The aqueous extracts of Citrus unshiu peel containing flavonoid glycosides was used as co‐substrate with Schizophyllum commune mycelia producing β‐glucosidase and its biological activities were studied. β‐glucosidase‐produced S. commune mycelia converted the glycosides (narirutin and hesperidin) into aglycones (naringenin and hesperetin).

Cells from the skin have been cultured for up to one month to detect the expression of keratins, actin, vimentin and extracellular matrix proteins (fibronectin, chondroitin sulphate proteoglycan, elastin and collagen I). Keratinocytes and dermal cells weakly immunoreact for Pan-Cytokeratin but not with the K17-antibody at the beginning of the cell culture when numerous keratin bundles are present in keratinocyte cytoplasm.
